Common acne drugs stop lung damage: A team of researchers from the Universities of Leeds and California, San Diego
have found that an antibiotic which is commonly used to treat acne can prevent
tissue damage caused by lung diseases such as emphysema.
The research involved studying a protein called VEGF, it was found that
doxycyline which is used to treat common ailments such as acne, sinusitis
and urinary tract infections - also boosts the body's ability to protect
against damage to the lungs.
VEGF helps to maintain healthy lung tissue and emphysema sufferers are found to
have unusually low levels of the protein. In healthy people, the lungs have an
active restorative system keeping them healthy, but in some lung diseases, the
body's natural protective processes are inhibited, partly as a result of low levels
of the VEGF protein.
In experiments, the tem of researchers reduced the levels of VEGF in the lungs
of mice whilst simultaneously administering doxycycline. The researchers found
that lung damage was minimal in these mice, compared with the control group
which was not treated with the drug. The images of the lungs of mice that
have been treated with doxycycline are startlingly different to those not
treated with doxycycline. VEGF-deficient lungs show vast pockets of tissue
damage when untreated and greatly reduced damage when treated with doxycycline.
While the experiments show that doxycycline clearly has a role to play in
preventing lung tissue damage, it is too early to say whether these findings
would have a preventative role for humans with a genetic predisposition to lung
disease.

Because familial emphysema results from a deficiency of AAT in affected
individuals, efforts to minimize the risk of emphysema have been directed at
increasing the circulating AAT levels either by promoting or increasing the
production of AAT within the individual, or augmenting it from the outside. One
strategy for improving the production of AAT is by pharmacological means (e.g.,
by administration of drugs such as danazol or estrogen/ progesterone

Methods to treat emphysema before it becomes disabling remain an important
research objective of programs supported by NHLBI. Since it is believed that
either excess protease (elastase), or too little useful antiprotease, can lead
to development of the disease, scientists have also been attempting to use other
approaches to develop animal models which will mimic the human condition of
inherited alpha-l-protease inhibitor deficiency and using such models to test if
natural or synthetic antiproteases can be used safely to prevent development of
emphysema-like lesions in these animals. If found safe and effective in animals,
these agents can be tried in humans.
