Medical Research Breakthroughs and Parkinson's Disease

Parkinson's disease in a new light: Two studies conducted at the University at Buffalo have shed new light on the development of Parkinson’s disease. The symptoms of Parkinson's disease (PD) result from disintegration of the brain's white matter, the network of nerves that transport messages to the various brain regions, and grey matter, the brain regions where those messages are received, interpreted and acted upon. The researchers used advanced magnetic resonance imaging (MRI) to identify brain regions linked to Parkinson's disease based on images. In of the studies the MRI scans and scores on the Mini-Mental State Examination (MMSE) of 155 patients diagnosed with early Parkinson's disease were compared with those of 101 normal subjects. Results showed that in Parkinson's patients, white matter hyper intensities [diseased areas of the white matter] were associated with significantly lower scores on the mental test: The more areas of hyper intensity, the lower the MMSE score. The second study investigated whether in early PD mild cognitive impairment is associated with a higher risk of developing dementia, by using a MRI analytical process called voxel-based morphology. Results in this study showed that mild cognitive impairment was associated with reduced grey matter in the cingulate area.The subjects in both studies were part of The Norwegian Park West project, a four-centre prospective longitudinal cohort study of patients with PD from south-western Norway. These studies are path breaking as they employ modern technology to explain facts which were erstwhile unknown.

Since alpha-synuclein fragments are implicated in both Parkinson and Alzheimer diseases, there may be shared pathogenic mechanisms between the two, therefore research into one disease may aid understanding of the other. Further avenues for research are also being suggested by cross-species comparisons assisted by database searching. Among others, rats, cows and zebra finches all possess alpha-synuclein; in the rat they play a role in the sense of smell, while in the zebra finch it is thought to be involved in the process of song learning. Further work to elucidate the function of alpha-synuclein in humans, and therefore clues as to the pathology of Parkinson disease, should be assisted by studying these other species.
